Brepocitinib meets all Phase 3 end points in noninfectious uveitis
Roivant and Priovant reported positive results from two Phase 3 studies; the indication has not been approved.
TL;DR
- Roivant and Priovant said brepocitinib met the primary endpoint in both CLARITY Phase 3 trials for noninfectious uveitis.
- The company-reported studies enrolled 166 and 204 participants, and treatment failure by week 48 was lower in the brepocitinib groups than in placebo groups.
- The results are topline company disclosures; regulatory review and full peer-reviewed reporting remain pending.
Roivant and its subsidiary Priovant reported positive topline results from CLARITY-1 and CLARITY-2, two Phase 3 trials of brepocitinib for noninfectious uveitis. The companies said the primary endpoint was met in both studies. [1] [2]
The sponsor reported that the trials enrolled 166 and 204 participants. At week 48, cumulative treatment failure was 35.4% with brepocitinib versus 78.7% with placebo in CLARITY-1, and 32.4% versus 77.8% in CLARITY-2. [1] [2]
The company also reported higher rates of steroid-free quiescence at week 48 in the treatment groups than in the placebo groups. These are sponsor-reported topline findings; the announcement does not substitute for a full peer-reviewed publication or regulator assessment. [1] [2]
Brepocitinib is already approved for dermatomyositis under the LISRAYA brand, according to the coverage. The noninfectious uveitis use remains investigational, and the companies said they intend to seek an additional indication in 2027. [1] [2] [3]
Why it matters
The results could support expansion of an existing drug into an inflammatory eye disease, but they remain a sponsor-reported clinical milestone. Regulatory review and detailed publication will determine how the topline efficacy and safety findings are interpreted.
